A Feedforward Mechanism Mediated by Mechanosensitive Ion Channel PIEZO1 and Tissue Mechanics Promotes Glioma Aggression

Alteration of tissue mechanical properties is a physical hallmark of solid tumors including gliomas. How tumor cells sense and regulate tissue mechanics is largely unknown. Here, we show that mechanosensitive ion channel Piezo regulates mitosis and tissue stiffness of Drosophila gliomas, but not non-transformed brains. PIEZO1 is overexpressed in aggressive human gliomas and its expression inversely correlates with patient survival. Deleting PIEZO1 suppresses the growth of glioblastoma stem cells, inhibits tumor development, and prolongs mouse survival. Focal mechanical force activates prominent PIEZO1-dependent currents from glioma cell processes, but not soma. PIEZO1 localizes at focal adhesions to activate integrin-FAK signaling, regulate extracellular matrix, and reinforce tissue stiffening. In turn, a stiffer mechanical microenvironment elevates PIEZO1 expression to promote glioma aggression. Therefore, glioma cells are mechanosensory in a PIEZO1-dependent manner, and targeting PIEZO1 represents a strategy to break the reciprocal, disease-aggravating feedforward circuit between tumor cell mechanotransduction and the aberrant tissue mechanics. Spatial resolution enhancement of coherent Doppler wind lidar using joint time–frequency analysis

Spatial resolution enhancement of a coherent detection Doppler wind lidar (CDWL) is generally achieved by reducing the pulse duration of the transmitted laser according to the basic lidar equation, inevitably, at sacrifice of other properties, such as the signal to noise ratio, maximum detection range and real time data processing. To solve this problem, a joint time–frequency analysis (JTFA) method is adopted in the CDWL. By comparing the performance of Cohen’s class in analysis and wind retrieval simulation, the adaptive optimized kernel (AOK) is picked up as the best JTFA method in the CDWL. In experiment, wind profiles retrieved using the JTFA and the FFT are compared with the result simulated using the software ANSYS 18.0. Benefiting from the high spatial resolution of the JTFA method, the dynamic structure of wind shear in the gap of two tall buildings is detected clearly, which agrees well with the simulation.

Diagnosis, treatment and long-term outcome of autoimmune pancreatitis in Sweden

Autoimmune pancreatitis (AIP) is a pancreatic inflammatory process characterized by a strong inflammatory cell infiltration and two histopathologically distinct subtypes: type 1 and type 2. Diagnosis is often challenging and requires a combination of clinical, laboratory and imaging data. AIP can mimic pancreatic tumours leading to unnecessary resections if not correctly diagnosed. Short- and long-term outcomes of AIP have been poorly investigated so far and no large series have been previously reported from Sweden.

Measurement of Singly Cabibbo-Suppressed Decays $D^{0}\to\pi^{0}\pi^{0}\pi^{0}$, $\pi^{0}\pi^{0}\eta$, $\pi^{0}\eta\eta$ and $\eta\eta\eta$

Using a data sample of $e^+e^-$ collision data corresponding to an integratedluminosity of 2.93 $fb^{-1}$ collected with the BESIII detector at acenter-of-mass energy of $\sqrt{s}= 3.773~GeV$,we search for the singlyCabibbo-suppressed decays $D^{0}\to\pi^{0}\pi^{0}\pi^{0}$,$\pi^{0}\pi^{0}\eta$, $\pi^{0}\eta\eta$ and $\eta\eta\eta$ using the double tagmethod. The absolute branching fractions are measured to be$\mathcal{B}(D^{0}\to\pi^{0}\pi^{0}\pi^{0}) = (2.0 \pm 0.4 \pm 0.3)\times10^{-4}$, $\mathcal{B}(D^{0}\to\pi^{0}\pi^{0}\eta) = (3.8 \pm 1.1 \pm0.7)\times 10^{-4}$ and $\mathcal{B}(D^{0}\to\pi^{0}\eta\eta) = (7.3 \pm 1.6\pm 1.5)\times 10^{-4}$ with the statistical significances of $4.8\sigma$,$3.8\sigma$ and $5.5\sigma$, respectively, where the first uncertainties arestatistical and the second ones systematic. No significant signal of$D^{0}\to\eta\eta\eta$ is found, and the upper limit on its decay branchingfraction is set to be $\mathcal{B}(D^{0}\to\eta\eta\eta) < 1.3 \times 10^{-4}$at the $90\%$ confidence level.

Observation of $\psi(3686) \to \eta' e^+ e^- $

Using a data sample of $448.1 \times 10^6$ $\psi(3686)$ events collected withthe BESIII detector at the BEPCII collider, we report the first observation ofthe electromagnetic Dalitz decay $\psi(3686) \to \eta' e^+ e^-$, withsignificances of 7.0$\sigma$ and 6.3$\sigma$ when reconstructing the $\eta'$meson via its decay modes $\eta'\to\gamma \pi^+ \pi^-$ and$\eta'\to\pi^+\pi^-\eta$ ($\eta \to \gamma\gamma$), respectively. The weightedaverage branching fraction is determined to be $\mathcal{B}(\psi(3686) \to\eta' e^+ e^-)= (1.90 \pm 0.25 \pm 0.11) \times 10^{-6}$, where the firstuncertainty is statistical and the second systematic.

Measurements of Absolute Branching Fractions for $\Lambda^+_c\to\Xi^0K^+$ and $\Xi(1530)^0K^+$

We report the first measurements of absolute branching fractions for the$W$-exchange-only processes $\Lambda^+_c\to\Xi^0K^+$ and$\Lambda^+_c\to\Xi(1530)^0K^+$ with the double-tag technique, by analyzing an$e^{+}e^{-}$ collision data sample, that corresponds to an integratedluminosity of 567 pb$^{-1}$ collected at a center-of-mass energy of 4.6 GeV bythe BESIII detector. The branching fractions are measured to be$\mathcal{B}(\Lambda^+_c\to\Xi^0K^+)=(5.90\pm0.86\pm0.39)\times10^{-3}$ and$\mathcal{B}(\Lambda^+_c\to\Xi(1530)^0K^+)=(5.02\pm0.99\pm0.31)\times10^{-3}$,where the first uncertainties are statistical and the second systematic. Ourresults are more precise than the previous relative measurements.
